CD4 is a single chain transmembrane glycoprotein expressed on the surface of T helper cells and regulatory T cells. CD4 is a co-receptor that assists the T cell receptor (TCR) with an antigen-presenting cell. CD4 interacts directly with MHC class II molecules on the surface of the antigen-presenting cell. CD8 is a transmembrane glycoprotein that serves as a co-receptor for the T cell receptor (TCR). The CD8 co-receptor is predominantly expressed on the surface of cytotoxic T cells, but can also be found on natural killer cells and dendritic cells. CD8 binds to a major histocompatibility complex (MHC) molecule, but is specific for the class I MHC protein. CD4-CD8 double staining reveals the distribution of T-lymphocyte subsets, for example for HIV infection, infiltrating cells in graft rejection and lymphoma.
